Sammendrag

Principal objective: Elucidate the genomic response to PPAR agonists and selective fatty acids on lipid metabolism in pig by transcript profiling and functional studies in vitro, particularly in relation to development, accumulation and breakdown of lipids in muscle and adipose tissue. Subgoals: 1) Elucidate the in vitro gene expression pattern during adipogenesis in porcine subcutaneous adipose tissue by use of high-throughput technology. 2) Determine the relative activation of the different PPARs by selective agonists and fatty acids by reporter gene studies in transfected cell lines. 3) Investigate the genomic response to PPAR-agonists and selective fatty acids in primary cultures of differentiated adipocytes and muscle. 4) Perform in vivogene expression pattern analysis of a selected number of genes.
