Tanytarsus hjulorum, new species, from a groundwater spring near Grahamstown, South Africa is described as adult male with associated partial COI gene sequences (DNA barcodes). The species is morphologically most similar to the species Tanytarsus luctuosus Freeman and T. usambarae Stur et Ekrem, but has clear diagnostic features in the male hypopygium and is easily separable from T.luctuosus and three other analyzed Tanytarsus species by its DNA-barcode. Tanytarsus elisabethae Ekrem, T. hjulorum and T. luctuosus were sympatric at the sampled locality in the Eastern Cape Province,while T. aterrimus Freeman was collected in the Cederberg Mts in the Western Cape Province.
